Abstract
The structure of sulfated o-linked oligosaccharides in glycoproteins was studied using MALDI-FTMS. O-linked oligosaccharides were released from jelly glycoproteins of X. laevis eggs by alkaline borohydride reduction. A new high performance liquid chromatography (HPLC) method was developed on a porous graphitized carbon column to separate the mixture into small pools containing one or few components. HPLC effluents were collected and analyzed by MALDI-FTMS in both positive and negative modes. The high mass accuracy of FTMS helped in the identification of four of the six reference compounds since their masses were the only ones observed.
