Isolation of mutants of cultured mammalian cells

Abstract

A very extensive review is given of the literature on mutation in cell cultures. The term mutation in this sense is clearly defined and experimental criteria are presented for this definition. The cell culture techniques required for isolation propagation and storage of mutant cell lines are described. Special attention is paid to the drug resistant mutants 8 azaguanine, 5 bromodeoxyuridine, ouabain and colchicine resistance. Moreover, the isolation of nutritional auxotrophs and conditional lethal temperature sensitive mutants is extensively discussed. (Bootsma - Rotterdam)
